Cell signalling over long distances makes use of endocrine cells, which secrete their signal molecules into the bloodstream, called hormones. The blood carries the molecules over long distances, allowing them to act on target cells that may lie anywhere in the body. Hormone binding to nuclear receptors induces conformational changes that accordingly alter the interactions of these receptors with other proteins in the cascade. Heterodimeric nuclear receptors for retinoids, vitamin D, and thyroid hormones are localized in the nucleus. In the absence of hormones with the corresponding response element, they repress the transcription of target genes. They activate transcription when bound to their ligands. Steroid hormone receptors are homodimeric nuclear receptors. In the absence of hormones, they are trapped in the cytoplasm by molecular chaperones. They can translocate to the nucleus and activate the transcription of target genes when bound to their ligands.
